Rat liver mitochondria accumulate iron mobilized from transferrin by pyrophosphate. The uptake has a very low energy dependence, but it is highly dependent on a functioning respiratory chain. Reduction of the ferric-iron-pyrophosphate complex is not linked to any specific respiratory complex. Half of the amount of iron accumulated is passed into heme. Iron once accumulated is very little accessible to chelation by added ferric or ferrous iron chelators. Iron uptake and heme synthesis are maximal if a suitable porphyrin substrate is added simultaneously with iron. The results represent further evidence that pyrophosphate is a possible candidate for intracellular iron transport. Also, the results suggest that iron uptake is coupled to simultaneous porphyrin uptake and heme synthesis.  相似文献

Treatment with interferon protected HeLa cells from infection with reovirus. This virus apparently activated an antiviral mechanism that was detected by the presence of (2'-5')oligoadenylate [(2'-5')An] in intact cells. The (2'-5')An was previously shown to activate an endoribonuclease, RNase L. We measured (2'-5')An by a sensitive competition-binding assay in cells infected at different multiplicities and for different lengths of time. Nanomolar concentrations of (2'-5')An were detected in cells infected at a multiplicity of greater than 5 after 2 h of infection, the time at which the infecting virions were uncoated. The level of (2'-5')An increased up to 6 h postinfection but declined afterward. To establish whether viral mRNAs were cleaved by RNase L, we analyzed the RNA extracted from infected cells by a highly specific hybridization assay on Northern blots. Full-sized reovirus mRNAs were detected in control infected cells, but not in interferon-treated infected cells, at 6 h postinfection. At this time, a nuclease activity could be detected in these cells by demonstration of cleavage of rRNA, degradation of cellular mRNA, and polysome breakdown in the presence of emetine. Since this inhibitor freezes ribosomes, cleavage of mRNA between ribosomes could only be accounted for by an endonuclease, presumably RNase L.  相似文献

Photorespiration by Chlamydomonas reinhardtii and Anacystis nidulans was measured as the oxygen inhibition of CO2 uptake and the CO2 compensation points. Net photosynthesis was oxygen dependent in Chlamydomonas grown in 5% CO2, but CO2 insensitive in cultures bubbled with air. Anacystis, even when cultured in 5% CO2, exhibited an CO2 insensitive net photosynthesis. The CO2 compensation point of Chlamydomonas grown in cultures bubbled with air and Anacystis grown in 5% CO2 enriched air, were reached shortly after the measurement was begun and the values were very low, less than 10 μl CO2 1?1; while Chlamydomonas grown in 5% CO2 enriched air for 4 days showed a high, but temporary CO2 compensation point (60 μl CO2 1?1). After a two hour adaptation in low CO2, a stable, low CO2 compensation point was reached. It seems that photorespiration can only be detected by the methods used in this study when the algae are cultured in high CO2, but a mechanism exists which blocks photorespiration when the green algae are adapted to low CO2 concentrations. When Chlamydomonas was treated with Diamox, an inhibitor of carbonic anhydrase, after cultivation in low CO2 (air), the cells behaved as if they had been grown in high CO2. They showed an oxygen sensitive net photosynthesis and a high CO2 compensation point. This indicates that carbonic anhydrase plays an important role in the regulation of a measurable photorespiration in Chlamydomonas. The results are discussed in relation to previous observations of photorespiration measured by enzyme assay, metabolic products and gas exchange properties.  相似文献

The differentiation of Trypanosoma cruzi epimastigotes into trypomastigotes was studied in diffusion chambers sub-cutaneously implanted in mice. Using epimastigotes of the Tulahuén strain, transformation was first evident at 16 h after implantation and reached its maximum (92% trypomastigotes) by 24 h. Shortly before their differentiation into trypomastigotes, epimastigotes were found to develop resistance to lysis by the alternative pathway of complement. Furthermore, implantation of stationary-phase (as opposed to log-phase) parasites resulted in the accumulation of large numbers of complement-resistant epimastigotes in the chambers. These observations suggest that epimastigotes pass through a complement-resistant transitional stage before differentiating into trypomastigotes and that transformation may require cell division. In a further series of experiments, epimastigotes recovered 7 h after implantation in mice were found to differentiate into trypomastigotes when cultured in vitro for an additional 17 h at 37°C. This observation indicates that the events which trigger the morphologic transformation of epimastigotes into trypomastigotes can be dissociated operationally from the differentiation process itself.  相似文献

Stem and leaf photosynthesis were measured in Glycine max var. essex (soybean) and Sparteum junceum (Spanish broom). The significance of stem photosynthesis to whole plant growth was evaluated by blocking stem photosynthesis with black straw sections. The growth of S. junceum was reduced by 18% when black straws were used in comparison to clear straws. The whole plant growth of G. max was not influenced by blocking the stem carbon contribution. Mean midday leaf photosynthesis was 12 μmol CO2 m–2 s–1 and 17 μmol CO2 m–2 s–1 for G. max and 5. junceum, respectively. Mean midday stem photosynthesis of S. junceum was 6.5 μmol CO2 m–2 s–1; however, positive net photosynthesis did not occur in G. max stems. Water stress caused a proportionally greater decrease in leaf photosynthesis compared to that of stems during diurnal cycles of photosynthesis in S. junceum. As a result the contribution to canopy carbon gain by stem photosynthesis increased from 38% to 48% of the total plant carbon gain under reduced water availability.  相似文献

Summary Two shoot populations of the rhizomatous, patchforming herb Solidago canadensis were studied throughout a developmental cycle in two abandoned pasture sites in southern Ontario. The shoot cohorts that emerged in spring dominated the two populations; subsequent recruitment was very low. Shoot mortality was highest in June and was concentrated in the smallest size classes. Both populations showed a pronounced bimodal size structure for most of the growth cycle. Relative growth rate of shoots declined as the growing season progressed and tended to be lowest in the smallest size classes. Inflorescence production depended on shoot size. The calculated relationship between log mean weight and log density of shoots was not constant during the growth cycle and the calculated maximum biomass values do not transgress the ultimate thinning line suggested from previously published data.Address for proofs and present address: Department of Geography, University of Liverpool, P.O. The effects of abscisic acid (ABA) on photosynthesis, dark respiration, and photorespiration were studied in Lemna gibba L. plants. The initial concentration of ABA in the nutrient solution was 10−7M and in a few experiments, 10−6M. The cultures were grown in the same solution for time periods ranging from one hour to 12 days. Net photosynthesis, measured as CO2 uptake by infrared gas analyser technique, was inhibited after four hours of ABA treatment and reached a minimum after four to seven days depending on the time of the year. After 12 days a substantial recovery of photosynthesis was observed. Dark respiration was significantly stimulated after two to seven days of ABA treatment but then returned to the control level. The transient effects of ABA on photosynthesis and dark respiration corresponded to the previously measured time course of [14C]-ABA uptake by Lemna . Photorespiration measured as oxygen inhibition of photosynthesis was not affected by ABA.  相似文献

17beta-[6,7- 3H]Estradiol was incubated with adult human liver slices in Krebs-Ringer phosphate buffer containing glucose. Of the identified 3H recovered, 51-76 percent consisted of estrone-3-sulfate (E13S) and 17 beta-estradiol-3-sulfate (E23S). E13S was the main metabolite and was found in both tissue and medium. E23S was present only in the medium. Minor amounts of estrogen glucuronides were formed. When a human liver homogenate was incubated with [3H]E2 in a medium fortified with excess uridine diphosphate glucuronic acid only some 4 percent of conjugation with glucuronic acid was observed. It is suggested that human liver favors sulfurylation as the conjugating mechanism for E2 and E1.  相似文献
